Abstract:<正> Recently a novel diterpene triepoxide-16-hydroxytriptolide-was discovered by researchers in theInstitute of Dematology of CAMS.This discovery followed a report of the active anti-inflammatory,im-munosuppressive and antifertility effects of another 5 monomers (T2,T3,T4,T6T28)isolated fromTripterygium wilfordii.
Abstract:A least.squares refinement procedure has been proposed for resolving enantiomorphous am- biguity of noncentrosymmetric structures containing heavy atoms in a centrosymmetric ar- rangement.During the least-squares refinement of a pseudo-centrosymmetric image containing both enantiomorphs, the temperature factors of atoms in one enantiomorph shift in the same direction, while those of the other shift in the opposite direction.Accordingly the true structure can be distinguished easily from its enantiomorph.Tests on four unknown struc- tures have shown that the method is very powerful.
Abstract:Human sperm membrane antigens extracted by deoxycholate (DOC) were used to immunize BALB/c mice.Hybrid cell lines secreting sperm-specific monoclonal antibodies were generated by cell fusion in a semi-solid medium and screened by indirect immunofluorescent assay using live and methanol-fixed sperm.Out of 850 hybrid clones from cell fusion,28 were shown to secrete sperm-specific antibodies which reacted with the acrosome, equatorial segment, whole surface plasma membrane or tail of spermatozoa. Finally, seven hybrid cell lines were established and shown to secrete monoclonal antibodies which had no cross-reactivity with arty human tissues other than testis and sperm. The majority were also shown to inhibit fertilization of mouse oocytes in vitro and human sperm penetration of zona-free hamster ova.Western blot analysis revealed that some of these antibodies reacted with sperm membrane antigens of distinct molecular size.
Abstract:A human B cell line (3D5) that responds specifically to B cell growth factor (BCGF) has been developed by a sequence of Staphylococcus aureus Cowen I activation,EB virus im- mortalization, and cloning.Proliferative response to PHA-stimulated T cell supernatant (PHA-T-Sup) and nonresponsiveness to rIL-2 stimulation were factors used to screen positive cells.Phenotype analysis with a flow cytometer indicated that:1) 3D5 is a B cell line: 100% of the cells were positive for B1 marker and 59% were positive for sIg,while T3 and Mo 1 were negative:2) 3D5 is an activated B cell line:both Tac and 4F2 markers of activated (but not of resting) B cells were 100% positive:3) 3D5 expresses high molecular weight BCGF (HMW-BCGF) receptor-associated epitope BA5.3D5 cells proliferated in response to cpBCGF stimulation in a dose-dependent manner.HMW-BCGF also induced 3D5 cells to proliferate.Interestingly.no proliferation could be detected in the presence of rIL-2,rIL-4,or rIFN-r.The data show that 3D5 cells are specifically BCGF-responsive B cells.Using 3D5 cells as target,BCGF activity was detected in crude BCGF preparation sedimented by 85% (NH4)2SO4 and chromatographed in a DEAE-Sephadex A-25 column from PHA-T-Sup.T24 cell supernatant with B cell differentiation factor (BCDF) activity could not induce 3D5 cells to differentiate into immunoglobulin-secreting cells.
Abstract:In recent years.diagnostic imaging techniques, especially ultrasonography (US) and CT scanning, have been widely adopted in clinical practice, making early accurate diagnosis of renal tumors possible.A total of 452 cases of renal tumors have been admitted to the institute since 1951,of which 220 were seen from 1951 to 1979 and 232 in the past 9 years (1980-1988).The frequency of renal parenchymal tumors was obviously higher in the latter group, including asymptomatic renal carcinoma in 20.2% and hamartoma in 38.1%. All these were discovered on routine physical check-up by ultrasonography and/or CT scanning and would otherwise have gone undiagnosed on conventional urography.Ultra- sonography and CT can also reveal the nature and the extent of the tumor. The idea that “a renal tumor should be considered malignant unless pathologically proven otherwise”is no longer valid. However, general manifestations of renal carcinoma, such as elevated erythrocyte sedimentation rate (ESR),hypertension, malaise, anemia, fever and hypercalcemia, still deserve proper attention. We suggest that ultrasonography of both kidneys should be mandatory in routine physical check-up,as far as the urinary system is concerned,in order to discover asymptomatic renal tumors.
Abstract:Thirty-one multiple myeloma patients were classified chiefly by heavy and light chain detec- tion of myeloma protein (M protein). The results were as follows:19 out of the 31 cases belonged to the IgG class, including the following subclasses—IgG1-λ.7;IgG1-k.5; IgG3-λ,1;IgG3-k,2;double M myeoma,2 (IgG3-λ+IgG2-k,IgG1-λ+IgG4-λ);IgG-λ,1;and IgG1-k 1.Seven cases were light chain diseases,of which 4 belonged to λ type.2 were k. and one was a double light chain disease.Five cases belonged to the IgA class (IgA-λ,3; IgA-k,2).We found no correlation between electrophoretic mobility and the antigenicity of M protein even within subclasses, and the highly concentrated BJP in urine had a tendency to form light chain polymers, possibly along different polymerization pathways.
Abstract:Sixty-six patients with advanced adenocarcinoma of the cardia underwent total gastrectomy between 1978 and 1986.The operative morbidity rate was 7.5% and the hospital mortality rate (1 month) was 1.5%.The postoperative 1,3,4,and 5 year survival rates were 69%. 50.7%,20% and 13.8% respectively. The quality of life after surgery was improved.Patho- logical type, lymph node metastasis, completeness of resectionand postoperative chemotherapy were the main factors influencing postoperative survival time.Indications for total gastrec- tomy and the advantages of transthoracic total gastrectomy are discussed.
Abstract:Fifteen cases,27 hips,of idiopathic necrosis of the femoral head (INFH) were included i our series. The role of radiography, isotopic bone scanning, intraosseous pressure measure- ment and intraosseous venography in the diagnosis of INFH were evaluated.Radiological examination could not reveal early cases, while isotopic bone scanning was more sensitive. There was an increase of intraosseous pressure in most of the diseased hips.and intraos- seous venography showed abnormalities in all examined hips. The latter two tests could therefore detect early INFH.Core decompression was performed in 23 hips,21 of which were followed for 7 to 27 months.Functional evaluation was good or excellent in 18 hips. Radiological re-examination remained unchanged in 19 hips.Postoperative intraosseous pressure measurement and intraosseous venography revealed a decrease of intraosseous pres- sure and improvement of venous drainage.Core decompression is therefore an ideal method of treatment for early INFH.
Abstract:Quantitative analysis of dendritic cells (DC’s) was carried out in tissue specimens of normal gastric mucosa (n=15),gastric ulcer (n=19),chronic atrophic gastritis (n=28),and gastric carcinoma (n=65) by ABC immunostaining with S100 protein antibody.Significant increase in DC number were observed in chronic atrophic gastritis with type Ⅲ intestinal metaplasia and/or grade Ⅱ,Ⅲ dysplasia. The result suggests that DC’s are potentially capable o presenting neoantigens associated with malignant transformation at the precancerous stage when malignant morphological changes have not yet taken place.Combined with routine diagnostic methods, the serial monitoring of DC density in gastric mucosa may be useful in the follow-up of premalignant lesions in the stomach and the diagnosis of early gastric carcinoma.
Abstract:The N-glycine constant infusion method was used to study protein turnover in normal adults and perioperative patients. The protein turnover rate (Q),synthetic rate (S) and catabolic rate (C) were derived according to Picou’s two-pool system model, and the“end-product averaging technique”showed the 15N-glycine constant iufusion method to be reliable, with a reproducibility of ±5.1%.This measurement can be repeated after a short period,so it should be useful for surgical nutrition study. Our results show the following:Q was significantly increased after operation (3.4± 0.6 to 5.0±0.5 g protein/kg/d,P<0.01),and S (2.6±0.5 to 3.7±0.6 g protein/kg/d,P<0.05) and C (2.3±0.5 to 3.9±0.5 g protein/kg/d,P<0.01) were increased as well. This suggests that the protein degradation rate outstripped the synthesis rate on post-operative day three (POD+3) when traditional (glutamine-free) total parenteral nutrition support was used.
Abstract:Myasthenia gravis is an autoimmune disease involving the acetylcholine receptors at neuromuscular junctions.Perioperative management of patients with myasthenia gravis is complicated by their enhanced sensitivity to nondepolarizing neuromuscular blocking agents and their resistance to depolarizing drugs (1).Prolonged respiratory depression commonly occurs when these agents are used.Atracurium dibesilate is one of a new series of nonde- polarizing agents and is promising by virtue of its rapid degradation by‘Hoffmann elimina- tion’.leaving products with little or no muscle blocking activity. After encouraging results were obtained with atracurium (2-6),we investigated the neuromuscular blocking effect of different doses of the drug (0.2 mg/kg,0.5 mg/kg) in sixteen myasthenic patients undergoing thymectomy.with continuous monitoring of neuromuscular function.
Abstract:Hyperthermia (42-44℃) and photosensitizing therapy can destroy S180 tumor cells, reduce malignant ascites and prolong the survival times of mice with carcinomas. The highest curative effect was observed when using a combination of the two treatments.Heating to 44℃ has a greater destructive effect on tumor cells than has heating to 42℃.The results show that this is due to a synergistic interaction between these two treatments. The fluores- cence spectrum of S180 cells was determined before and alter treatment, and the indication was that the synergistic effect is probably related to a new fluorescence product;the greater the intensity of the new fluorescence、the more marked the synergy of hyperthermia and photosensitizing therapy. The maximum emission wavelength was 460nm (excitation wave- length 370nm).
Abstract:The differences of serum lipid and lipoprotein (LP,profiles of animals susceptible (rabbit) and nonsusceptible (Beijing duck) to atherosclerosis as well as the distribution of apolipo- protein (apo) A-I and its catabolism in vivo were studied.Eight items,i.e.total cholesterol (TC),high density lipoprotein (HDL) cholesterol, triglyceride, percentage of a-LP and β-LP, β-LP concentration, lecithin cholesterol acyl transferase (LCAT),and agarose and polyacryla- mide gel electrophoresis of both species were assayed before and after feeding a high fat, high cholesterol diet. Results indicate that the exogenous cholesterol consumed by Beijing ducks was carried and transported by HDL,while that in rabbits was transported by low density lipoprotein (LDL).The biological half lives of apo A-I in serum and in HDL were 41.11±2.4 and 42.8±1.7 h respectively.and its distribution in different organs was in the order of liver>kidney>spleen>lung>heart>intestine>muscles>aorta. These results show that the liver is the major organ for metabolizing HDL apo A-I.and the kidney is also important. The results also imply that in Beijing ducks the cholesterol carried by HDL may be catabolized through apo A-I receptors in the liver and kidney. The differences in cholesterol binding and in lipoprotein and apolipoprotein metabolism of the two species provide important clues for the elucidation of the pathogenesis of atherosclerosis.
