Abstract:Glycidyl methacrylate (GMA)is a recently recognized mutagen. In order to explore the mutagenicity and mechanism of GMA, plasmid PBR322 was used for in vitro binding, mutant screening, restriction enzyme mapping, and DNA sequencing. To explore the mechamism by which an initial premutational event is converted into a stable heritable mutation, pBR322 and GMA-bound pBR322 were transformed into E. coli HB101 , and the following results were obtained : 1) GMA-bound PBR322 induced phenotype changes in competent cells. Two stable and heritable mutants were isolated (Ap ̄RTc ̄S and Ap ̄STc ̄R). 2) When restriction enzyme mapping was used to analyze the mutant Ap ̄RTc ̄S , four of seven maps showed changes, but no large DNA insertion or deletion were observed.3) The frequency of deletion and insertion forms counted about 10%. Sequence specificity and hot spot regions were evident in the sequence analysis of mutated plasmid. The above results indicate that the premutagenic lesions of plasmid induced by GMA can be converted into point mutations in vivo.
Abstract:Iontophoresis of hydrogen ions (H ̄+ ) to respiration related units (RRUs) in the nucleus retrofacialis (NRF)region was used to investigate the role of RRUs in central chemoreception of respiration. The results were as follows:1) of the 1 3 inspiration related units (InRUs) recorded , 7 were excitatory and 1 was inhibitory; 2) of the 29 expiration related units (ExRUs). 12 were inhibitory and 1 was excitatory; and 3) of the 7 phase-spanning respiration related units (PSUs). 4 were excitatory and 2 were inhibitory. None of the remainder were affected.Histochemical study using the horseradish peroxidase (HRP) technique revealed a neural structural relation between the ventral surface of the medulla and neurons in the NRF region. It was suggested that the RRUs in the NRF region might contribute to the central chemoreception which regulates respiration.
Abstract:This paper reports that lesions of the medial frontal cortex (MFC) caused behavioral deficit in rats and that this deficit could be attenuated by a well formulated treatment with Org2766, an analog of ACTH- (4-9). Wistar rats were distributed in 3 groups: MFC lesion treated with saline (M-N); MFC lesion treated with Org2766 (MO);and sham-operation treated with saline (S). Repeated Org2766 or saline injection commenced from the day of surgery and lasted for 1 3 days. After surgery. the rats were trained in a passive avoidance task and then in an active avoidance task. MFC lesions were found to be strongly associated with behavioral deficits. The M-N group rats displayed poor retention of the passive avoidance response and showed much slower learning of the active avoidance task as compared to S group rats. The result showed that chronic Org2766 administration improved the behavioral performance of both tasks in MFC lesioned rats. The also revealed that the superoxide dismutase (SOD)activity was significantly increased in the M-O group as compared to the M-N group 15 days after surgery. The possible mechanisms related to the beneficial effect of Org2766 on cortex damage are discussed.
Abstract:Proliferative activity in 106 cases of non-Hodgkin’s lymphoma was estimated using the monoclonal antibody Ki67 and by counting the number of mitotic figures. The percentage of Ki67-positive cells was compared with the median number of mitotic figures per square millimeter. Both Ki67 positivity and the number of mitotic figures were found to be greater in high grade than in low grade lymphomas, although there was an overlap between the two grades of malignancy. A close correlation was found between the number of mitotic figures and the percentage of Ki67-positive cells not only in all lymphoma typo taken together (rs= 0. 834 . P<0. 001). but also in B-cell lymphoma (rs= 0. 8 1 P< 0. 001) and T-cell lymphoma (rs = 0. 764. P<0. 00 1) taken seprately. Thus, both methods are useful for the estimation of proliferative activity. but each method has its advantages and disadvantages.
Abstract:In the past few years. a total of 6 different mutations of the G6PD gene have been reported in china. One of these, the C6 mutation (A ̄(95)→G). accounted for about 1 5. 4 % of the Chinese G6PD variants. In ordet to develop a strategy for rapid detection of mutation-containing exons of the G6PD gene. we applied the single-strand conformation polymorphism (SSCP) technique to the detection of mutations in exon 2 of this gene. We observed four patients with abnormal migration patterns of the exon 2 bend among 20 cases of G6PD variants. Direct PCR sequencing confirmed a T to C substitution in exon 2 that has previously been reported. This procedure is therefore of particular importance for the rapid detection of mutation-containing exons in the G6PD gene.
Abstract:Expression of c-myc oncogene transcripts in colorectal neoplasia was studied in paraffin embedded tissue sections from 25 patients undergoing surgery and from the rectal carcinoma cell line HR-8348 by using in situ hybridization, and its amplification was investigated in tumor and normal mucosa tissue from 25 coloproctomy samples by slot blot hybridization. Overexpression of this gene was seen in 78% (7/9) of the benign adenomas and 91% (20/22) of the malignancies sampled. There was no significant correlation between overexpression and the histologic type or grade, and no significant relationship between the level of expression and clinical stage was found, although overexpression was apparently more common in tumors with metastasis. Amplification of the gene was found in 0 of 4 benign adenomas and 7 of 22 malignancies. No obvious correlation was found between amplification and histological type or grade, though amplification was more frequent in tumors with metastasis. Amplification was also found in 2 adenomas with malignant change. The results suggest that multiple factors are involved in the progression of colorectal cancer , and in situ hybridization with a nonradiolabeled probe is useful in the detection of gene expression.
Abstract:We studied 14 moderately overweight Typo 2 diabetic patients with essential hypertension in stable metabolic control after a run-in period , and again after 3 months of antihypertensive treatment with the angiotensin-converting enzyme (ACF) inhibitor captopril. Glucose tolerance was tested with a 75g oral glucose load (OGTT) and insulin sensitivity was measured by the insulin suppression test (IST) while dietary and drug treatment of the hyperglycemia was maintained constant. In the whole group. mean blood pressure (MBP) fell progressively over 3months from a baseline value of 123± 3 mmHg (1 mmHg= 0. 133 kpa) to a final value of 115± 2 mmHg(P<0. 005). After treatment, fasting plasma glucose, insulin, free fatty acid (FFA). potassium, and glycosylated hemoglobin concentrations were unchanged from baseline. There were no significant differences in glucose tolerance and insulin sensitivity between pre- and post-trearment values. Neither endogenous (oral glucose) nor exogenous (IST) insulin caused any change in plasma potassium concentration. This resistance to the hypokalemic action of insulin was not affected by captopril.
Abstract:Early pregnancy factor (EPF) was purified from the pooled sera of 21 0 pregnant women at 3- 8 weeks of gestation. Sera from healthy nonpregnant women were used as control. The samples (G-Ⅱ . G-Ⅲ and G-Ⅳ) obtained from pregnant women had EPF activity but no HCG activity. Polyacrylamide gel electrophoresis showed that the major bands in pregnant G-Ⅲ and G-Ⅳ were at similar positions in tube gels. The results of SDS-PAGE showed 3 bands in pregnant G-Ⅳ: 57. 0 kD. 38. 0 kD and 19. 0 kD. The basic active form of EPF may be a small peptide of 1 9. 0 kD. The isoelectric points of pregnant G-Ⅳ were 6. 45 and 8. 20.
Abstract:The blood supply system of the optic chiasma was studied in 85 fresh human specimens using various histological and anatomical methods. Computer image analysis and ultrastructural examinations of the microvessel in 58fetal specimens were also conducted. The authors found that the medial portion of the chiasma is a weak point in the microcirculation network. This weak point is apt to be disturbed first and become ischemic. causing disorders of the crossing optic nerve fibers and resulting in characterestic bitemporal visual field defects. SEM studies showed no ultrastructural difference between the capillaries at the medial and lateral portions of the chiasma. It was concluded that: 1) No special artery supplies the median chiasma the weak point of microcirculation at the median chiasma is due to its relatively scanty capillary distribution; 2) "lateral chiasma arteries" could provide a better blood supply to the lateral fibers and thus the nasal quadrantic visual field could be preserved in many late stages of visual field defect in sellar region tumors, 3) cases with pituitary microadenoma which is not sufficiently large to press the chiasma but involves bitemporal visual field defect are due to the tumor recieving “shunt-flow” (stealing blood) from the chiasma through the peri-infundibulum plexus.
Abstract:In order to assess the role of tumor necrosis factor (TNF) in neonatal sepsis, plasma TNF levels were determined by a method using L929 cells at the time of septic work-up in 67 neonates. Thirty-three patients, with sepsis were found to have significantly higher TNF levels (533. 33 ±468. 74 U/ml ; 1 U corresponding to 1. 67 pg recombinant TNF) as compared with 34 non-sepsis patients (100. 0±188, 97 U/ml) and 30 healthy newborns (27. 33±1 6. 1 7 U/ml, P<0. 05. respectively) . The upper limit of normal plasma TNF levels was 60 U/ml and the best cutoff value for predicting neonatal sepsis was 160 U/ml. This had remarkable sensitivity (88%), specificity (82%). positive predictive value (83%). and negative predictive value (88%). Plasma TNF levels were significantly associated with the occurrence of shock, organ failure. sclerema and outcome. Thus, anti-TNF antibodies might be used in protecting newborns from septic death.
Abstract:Eighty-one patients with suspected renovascular hypertension (RVH) were studied using 99m Tc-MAG3 renography before and after administration of 25 mg captopril (CAP). Among the 81 cases, 22 were proven by renal arteriography or DSA. The results showed that by using CAP, the sensitivity and specificity of renography in RVH diagnosis increased from 66% to 72% and from 33% to 100%, respectively. We conclude that renography is a simple, noninvasive . sensitive and specific method for screening RVH and is suitable for use in hospitals with no Y-cameras.
Abstract:Prostaglandin E2 (PGE2) levels in gingival crevicular fluid (GCF) of 46 normal controls and 90 patients suffering from periodontitis with different periodontal pocket depths were measured by radioimmunoassay (RLA).The results demonstrated that PGE2 levels in the GCF of the periodontal pockets are higher in patients with periodontitis. The PGE2 level rises as the periodontal pocket deepens, especially in cases where the periodontal pocket depth exceeds 6 mm. This study shows that the PGE2 level is significantly related to the severity of bone destruction in periodontitis.
Abstract:Myasthenia gravis patients are hypersensitive to nondepolarizing relaxants, such as alcuronium, an intermediate-long nondepolarizing agent. This study observed the effects of alcuronium treatment in myasthenia gravis patients as compared with non-MG patients during operation. Ten MG patients (Ossermann class I - IV, scheduled for thymectomy) and 10 non-MG patients (ASA class I-Ⅱ, scheduled for operation) were selected. An induction dose of alcuronium 0. 2 mg/kg and thiopental 4-6 mg/kg was given , followed by intubation and ventilation with 50% nitrous oxide in oxygen and 0.5-1. 5% ethrane. Neuromuscular transmission was monitored using an accelerogragh and degrees of neuromuscular function at different depths were recorded. There were statistically significant differences between the two groups. The effect of alloferin in the MG group was quicker and deeper. This study also found a relation between MG class and the recovery of respiration: Respiratory recovery was quicker in classes I-Ⅱthan in classes Ⅲ-Ⅳ.
