Abstract:Ultrastructural immunocytochemical characterization of human long-term bone marrow cultures has shown positive localization for growth factors on cell surface and on extracellular matrix(ECM).In some cases double-labelling indicated co-localization of growth factors and specific cell surface labels.Specific markers for endothelial cells and fibroblasts showed that growth factor(GM-CSF,G-CSF and b-FGF) were present at the surface of these cell types. Both scanning and transmission electron microscopy indicated intense labelling for growth factors on the extracellular matrix.Double-labelling of heparan sulphate proteoglycans and GM-CSF showed a co-localization of the labelling, which indicated the binding of growth factor to the extracellular matrix.
Abstract:Eight different geographical districts in China varying in urban and rural characteristics from 6 provinces and 2 municipalities were selected as pilot areas of the project. The interventions included early detection and treatment of neuritis,self-care training, adapted footwear, surgery, comprehensive treatment of complicated ulcers, and prostheses.Main changes in eye, hand and foot impairments between baseline assessment and assessment at 2 years have been presented in this paper.
Abstract:Retinoids mediate their actions via retinoic acid receptors(RARs) and retinoid X receptors(RXRs).Each of class of these nuclear retinoid receptor is further subdivided into three species namelyα,βandγ.Recently studies demonstrated that estrogen receptor(ER)-positive human breast cancer(HBC) cell lines are sensitive and ER-negative cell lines are resistant to growth inhibitory effects of retinoic acid(RA).In this study,we found that only RARαmRNA levels was strongly correlated with ER-status.To further investigate the major role of RARαin retinoid-mediated inhibition of growth,we transfected RARαcDNA into two RA-resistant ER-negative HBC cell lines.Analysis of different clonal populations of RARαtransfectants from each cell line revealed growth inhibition by retinoids. Our results demonstrated that RARαplays a major role in mediating retinoids inhibition of growth in HBC cells and adequate levels of RARαare required for such an effect.
Abstract:DNA ploidy of 57 gastric carcinomas with metastases(12 liver,1 adrenal,4 ovary and 48 lymph node) were measured by flow cytometry.DNA anueploidy was significantly related to liver metastases:9 out of 12 gastric carcinomas with liver metastases were anueploid(75%) as compared to 13 out of 45(28.8%) of cases without liver metastases(P<0.01);the one gastric carcinoma with adrenal metastasis was also anueploid.DNA ploidy was not related to ovarian or lymph node metastases.Another interesting finding was that all of 3 gastric carcinomas with liver metastases which showed a diploid DNA pattern, expressed p53 protein, while all of 3 carcinomas with liver metastases but no p53 protein expression were anueploid. The expression of p53 protein was not related to ovarian metastases. The results suggested that an anueploid DNA pattern and the expression of p53 protein are both objective markers valuable in predicting high risk potential of metastases to the liver, and that the combined detection of these markers can be a most useful method in the follow-up of Patients with gastric carcinoma in detecting those at high risk of developing metastases following surgical resection. Also the poorer prognosis of Patients with gastric carcinoma showing an anueploid DNA pattern may be related to the development of distant organ metastases through the blood vascular system.Furthermore, the clone of gastric carcinoma cells which accumulate p53protein or show an anueploid DNA pattern may have a causative role in the development of liver(&.adrenal) metastases.
Abstract:The role of growth factors and proto-oncogene in pulmonary vascular structural remodelling is not well known. The present study examined gene expression of platelet-derived growth factor(PDGF)-A and -B chain and proto-oncogene,c-myc,in lung tissue and pulmonary artery of rats exposed to hypoxia and compared to those levels of gene expression in normal rats.Normal lungs and pulmonary artery expressed PDGF-A chain transcript of 1.7 kb and PDGF-B chain transcript of 3.5 Kb.The c-myc transcript of 2.2 kb was expressed as well. After hypoxic exposure for 7 and 14 days mRNA levels of PDGF-B chain and cmyc were elevated significantly compared with those of control rats.PDGF-A chain mRNA increased after hypoxia for 7 days, and then declined. These results suggest that activation of autocrine and/or paracrine is important in proliferation mechanism of pulmonary artery smooth muscle cells in hypoxic pulmonary hypertensive rats.
Abstract:The polymorphisms(Pvu Ⅱand Hind Ⅲ) on the lipoprotein lipase(LPL) gene locus was investigated in a sample of 100 patients surviving previous myocardial infarction and 100 age matched healthy individuals selected from Han Chinese of Beijing area.In patient group a strong association was found between H+allele of Hind Ⅲ polymorphism and raised TG levels(P<0.01).In control group P-P-genotype was observed to be associated with higher TG levels compared with P+P genotype of Pvu Ⅱ polymorphism(P<0.05).Combination of H+H+ genotype with P-P-genotype showed the highest TG levels among all nine kinds of genotype combinations in patient group(P<0.01).However,comparison of distribution of alleles and genotypes of these polymorphisms between patient group and control group demonstrated no significant difference. Our data suggest that the polymorphisms at the LPL gene,as the linkage markers with an aetiologic mutation at or around LPL gene, may constitute one of the genetic determinants for the population variation in plasma TG levels,as well as for the common dyslipidemia in Chinese population.
Abstract:The mutation of p53 gene was detected in Syrian hamster embryo(SHE) cells neoplastically initiated with a particles. The level of the p53 mRNA in transformant was obviously higher than that in non-irradiated counterpart,as measured by Northern blot analysis of total RNA.A pair of primers were designed based on p53 cDNA sequence to produce the whole length of coding sequence about 1.2 kilobase(Kb) by reverse transcription of mRNA followed by the polymerase chain reaction(RTPCR),but the length of fragment amplified from transformant mRNA was about 0.3Kb,remarkably shorter than that from normal SHE cells.Imrnunohistochemical analysis of p53 protein showed that no heavy staining was found on slice of tumor derived from transformant inoculated in nude mice with hamster specific p53 monocloned antibody HD200.The results implied that p53 gene had been mutated by deletion, which might lead to loss of p53 protein expression but the increased expression of p53 remained in a particle-induced SHE transformant.
Abstract:We used a potent and specific monoclonal antibody to somatostatin to test the physiologic inhibitory role of the tetradecapeptide somatostatin on pancreatic secretion.Somatostatin immunoneutralization increased both the total amylase and volume of pancreatic secretion.Cholecystokinin-A receptor antagonism abolished the stimulatory effect of somatostatin immunoneutralization. We conclude that somatostatin tonically inhibits pancreatic secretion in fasted rats via inhibition of the release or action of cholecystokinin.Furthermore, the source of these peptides is likely islet delta cells and intrapancreatic neurons, respectively.
Abstract:In this study,we investigated the effect of captopril(CPT) on glomerular filtration rate(GFR),effective renal plasma flow(ERPF),filtration fraction(FF),urinary albumin excretion(UAE) and daily urinary excretion of thromboxane B2(TXB2) and 6-keto-prostaglandin F1a(6-keto-PGFla) in 29 normotensive non-insulin-dependent diabetes(NIDDM) patients without clinically discernible nephropathy. Before treatment, urinary excretion 6-keto-PGF1a was significantly increased(P<0.05) in 29 NIDDM patients compared with 25 health subjects matched for age and sex.The values of GFR and FF were significantly higher(P<0.01 and P<0.005,respectively) in NIDDM than in normal volunters, whereas ERPF was comparable in both groups.Meanwhile we observed that UAE of early NIDDM was increased before treatment. After CPT treatment,GFR,FF,UAE and urinary excretion of 6-keto-PGFla were significantly reduce(all P<0.005) compared with those of NIDDM before treatment. These data indicated that CPT is effective in lowering glomerular filtration pressure and ameliorating microalbuminuria in the normotensive early NIDDM.

Abstract:One hundred and twenty-four patients with heart disease(75 cases of rheumatic heart disease,26 cases of congenital heart disease,13 cases of aortic disease and 10 cases of other disease)were examined by Omniplane transesophageal echocardiography(TEE).The result showed that Omniplane TEE transducer can be rotated from 0°to 180°in probe and had the advantages of broader scope, obtaining more information, less stimulation to esophagus and easy to manipulate. It suggests that Omniplane TEE is a efficient technique in clinical diagnosis and can be extensively used in the future.
Abstract:It is well known that serum immunosuppressive factors play an important role in the mechanism of postburn immunosuppression. This study was intended to investigate the effect of haptoglobin, purified from the serum of burned patients by affinity chromatography,on the proliferation and interleukin-2(IL-2) secretion of normal murine thymocytes induced by ConA and the proliferation of IL-2 dependent cell line(CTLL-2) stimulated by recombinant human IL-2,so as to elucidate the role of serum haptoglobin in postburn T-lymphocyte dysfunction. The results showed that purified haptoglobin,at the level equivalent to the concentration found in serum of burned patients, significantly inhibited the proliferation and IL-2 secretion of normal murine thymocytes as well as CTLL-2 proliferation;whereas it exhibited no immunosuppressive effects at the level equivalent to the concentration found in serum of nomal volunteers.According to the results reported here,it is suggested that extraordinary increase in serum haptoglobin level may be an important factor of impaired T-lymphocyte responses following burns.
Abstract:With the advance of spinal surgery in the last decade, surgical treatment of spinal tumors has been no longer limited to simple laminectomy. The principles of surgical treatment of spinal tumors include:(1) anterior approach for the anterior lesion and posterior approach for the posterior lesion;(2) combined anterior and posterior approach for extensive lesions and(3) internal fixation for spinal stability.32 cases of spinal tumors were treated on the basis of the above guiding principles and 84%(27/32) showed excellent or good results.
Abstract:Pulsed field gel electrophoresis(PFGE) has been firstly introduced in characterization of the pathogenic fungi Penicillium marne f fei and Exophiala dermatitidis genomes. The numbers and sizes of their chromosomes have been detected.Polymorphism was identified on the smallest chromosome of E.dermatitidis.The result shows that PFGE for characterization of large molecular DNA pathogenic fungi is very suitable,it is more simple and more efficacy. The result also shows the diversity of pathogenic fungi is relative common even in rare occurred pathogenic fungi such as E.dermatitidis.
Abstract:Three cases of pulmonary leiomyosarcoma were presented. The characteristic clinical features were described with review of literature. In comparison with bronchogenic carcinoma, the leiomyosarcoma has some characteristics:1) On chest X-ray,it usually appears as a sharply demarcated, even density round mass, growing rapidly within the lung, it rarely accompanies with hilar or mediastinal lymph node metastasis.2) The preoperative cytological or pathological diagnosis is difficult either by sputum smear or by bronchoscopic biopsy or by fine needle percutaneous aspiration biopsy.31 Pathological differential diagnosis of leiomyosarcoma of lung from anaplastic lung cancer is difficult. In conclusion, the primary pulmonary leiomyosarcoma is a rare malignant tumor, detecting the present illness seriously, paying attention to the chest X-ray films characterize, early surgical resection is the only way to get diagnosis and effective treatment method.
