Abstract:The expression of mRNA for bone morphogenetic protein-1 (BMP-1) was investigated in fetal human cartilage and bone during the developing endochondral ossification by using in situ hybridization. BMP-1 mRNA was localized in the hypertrophic chondrocytes, the cells of perichondrium, osteoblasts and marrow cells. The results suggest that BMP-1 may involve in cartilage and bone formation, and in regulation of calcification of growth plate. As a comparative study, the expression of TGF-β1 protein was investigated in cartilage and bone using immunohistochemical staining, correlated expression of TGF-β1 and BMP-1 was found. The results suggest that BMP-1 and TGF-β1 act synergistically to influence endochrondral ossification.

Abstract:Objectives. To study the expression of TNF α mRNA and the effect of somatostatin on the expression of TNF α mRNA in multiorgans of rats with acute hemorrhagic necrotic pancreatitis(AHNP). Methods.AHNP in the rat was induced by retrograde injection of 5% sodium taurocholate into the bile-pancreatic duct. Somatostatin octapeptide (SS-OP) (2μg/kg)was injected into the femoral vein imme- diately in rats of the treatment group after inductive AHNP. Rats of the sham operative group received in- jection of saline. Sixty animals of the AHNP and sham operative groups at the designated time(0. 2h, 0. 5 h, 2h, 4h, 8h, after the operation, six animals at each time point)and tweleve animals of treatment group at 4h after the operation were sacrificed for samples of pancreas, liver and lung. The expressions of TNF α mRNA within the pancreas, liver and lung were established by RT-PCR. Results. TNF α mRNA became detectable in the pancreas as early as 0. 2h after inductive AHNP, while it was undetectable in other organs until 0. 5h. Expression of TNF α mRNA in each tissue increased continuously and reached a peak at 4h,demonstrating a significant difference compared with that at 0. 5h and 8h. Expressions of TNF α mRNA from pancreas, liver and lung were decreased 50-80% in the treat- ment group, the pancreatic necrosis was also attenuated dramatically. Conclusion. TNF α mRNA was detectable in pancreas, liver and lung tissues at the early stage of AH- NP.SS-OP can significantly inhibit the expression of TNF α mRNA and attenuate the pancreatic necrosis. We feel that this may be an important mechanism of SS-OP in the treatment of AHNP.

Abstract:Objective. To investigate the relationship between mdrl gene expression and multidrug-resistance in o- varian carcinoma. Design. We established tumor-bearing mice model of ovarian carcinoma, compared the anticancer drug sensitivity of OC/mdr-cell and OC/mdr+ cell in vitro, investigated the effect of cyclosporin A on reversing the multidrug resistance both in vitro and in the tumor-bearing mice model, detected the mdrl gene expre- sion in human ovarian carcinoma specimens. Main outcome measures. Anticancer drug sensitivity of both OC/mdr-cell and OC/mdr+ cell is mea- sured by the methods of MTT assays. mdrl gene expression is detected by the methods of RT-PCR. Results. Using MTT assay,OC/mdr+ cell is 4. 1 - 15. 5 times more resistant to VP-16,VCR,DNR, and DOX than OC/mdr-cell in vitro.2 μg/ml cyclosporine A(CsA)reduced the resistance of OC/mdr+ cell to DOX,from 0. 324±0. 072μg/ml to 0. 088±0. 024μg/ml. To OC/mdr-cell,CsA did not significantly in- crease its sensitivity to DOX.Tumor-bearing mice with positive mdrl gene expression showed non-respon- siveness to DOX chemotherapy. When combined with intraperitoneal injection of CsA, the growth rate of tumor cells decreased significantly (P<0. 01 ). Only 4 of 23(17. 39% )tumors from patients who had not received chemotherapy exhibited positive mdrl gene expreession, while 6 of 9 (66. 67%)treated patients showed positive mdrl gene expression (Fisher exact test: P<0. 05 ). After cytoreductive surgery and chemotherapy, 14 of 19 untreated patients with negative mdrl gene expression had partial or complete re- sponse, while in patients with positive mdrl gene expression, 8 of 10 showed poor prognosis(Fisher exact test: P<0. 05 ). Conclusion. The expression of mdrl gene is associated with previous chemotherapy. CsA can reverse the resistance of mdr+ cells to indr-associated drs both in vitro and in vivo’ For the patients with ovarian carcinoma., the percentage of nonresponsiveness to the chemotherapy was found to be significantly higher among patients with positive mdrl gene expression than those with negative mdrl gene expression. mdrl gene expression can be detected to predict the clinical prognosis of patients with ovarian carcinoma.

Abstract:By southern hybridization with 1. 8 kb cDNA probe,a high frequency (40. 5 % ) of structural abnor- mality of p53 gene was observed in primary nasopharyngeal carcinoma (NPC) biopsies. The regions of ex- ons 1 to 4 of the gene were examined by polymerase chain reaction-single strand con formation polymor- phism,no point mutation was found. Because very low rate of point mutation had been reported in exons 5 to 8, we considered that structural abnormality in the region of e-cons 1 to 8 of the gene might be uncom- mon in NPC. The spectrophotometer scanning analysis of autoradiograms and rehybridization investigation of nitrocellulose filter with exon 11 probe indicated that most of structure aberrations we observed might be rearrangement occurring in exon 11.

Abstract:The object of this study is to investigate the properties of gastric cancer assoiciated antigen by McAb3H11 against gastric cancer cell line. Antigen purified by affinity chromatography and further charac- terized by biochemical and immunological techiques. Results showed that assoiciated antigen of McAb 3H11 was mainly located on the membrane of target cells. It was sensitive to heat and digestion of proteases, but resistant to treatment with sodium periodate. Schiff’s reagent staining was negative. SDS-PAGE and IEF- analysis showed that the antigen was a 210 kD molecular weight with pl of 8. 5 protein. The molecular weight of antigen extracted from target cell with tunicamycin was not changed. We concluded that the cor- responding antigen of McAb 3H11 might be an alkaline protein.

Abstract:Retinoic acid receptor-α (RAR α) plays a major role in the growth inhibitory effect of retinoic acid on human breast cancer cells, may be it could serve as an indicator to guide the treatment and prevent of breast cancer with retinoic acid in clinic. All previous researchs were based on observing the changes of RAR α mRAN expression. In this study, the expression of RAR α in human breast cell lines was studied by Northern Blot, Western Blot and Immunohistochemistry in mRNA level and protein level. Results showed that RAR α protein expression was correlated with RAR α mRNA expression. RAR α mRNA ex- pression was higher in estrogen receptor (ER)-positive human breast cancer cell lines than in ER-negative ones. So was RAR a protein expression. Both RAR α mRNA amd RAR α protein expression were associ- ated with ER status. The expression of RAR α and the relationship between RAR a and ER status were al- so determined by immunohistochemistry in 58 human primary breast cancer tumors. 37 (63. 8% ) tumors were ER-positive and of these 28 (75. 7% ) were also RAR α -positive. The coexpression of ER and RAR a was statistically significant (P<0. 01, by X2 contingency analysis). It was reported that RAR α expres- sion in cultured breast cancer cells was regulated by estrogen acting via the ER. Our study demonstrated that RAR α expression may be modulated in breast cancer in vivo by estrogen via ER.

Abstract:Purpose. To clarify the role of perforin-and Fas ligand (L)-mediated cytotoxicity pathogenesis of viral myocarditis. Materials and methods. Forty balb/c mice were randomly divided into experimental group (n = 20) and control group (n = 20), and inoculated intraperitoneally with coxsackievirus B3(CVB3) and Eagle’s solu- tion without CVB3, respectively. The mice were sacrificed and their hearts were removed at day 7 post-in- oculation. Expression of perform and FasL were detected with immunohistochemistry, reverse transcription-polymerase chain reaction (RT-PCR) and in situ hybridization. Results. (1)Perform-and FasL-positiye cells were demonstrated in experimental murine hearts by im- munohistochemistry, however, no cells were discovered in control murine hearts; (2) The examination of RT-PCR showed the positive ratios of perform and FasL mRNA in myocardium were significantly higher in experimental group (100% and 100 % ) than that in control group (20% and 30 %, P<0.05); (3)Positive signals of perform and FasL mRNA were found in myocardium of all the experimental mice by in situ hybridization, but nothing was detected in control group. Conclusion. Perform and FasL can be expressed in infiltrating cells in murine myocardium with acute myocarditis caused by CVB3, suggesting perform and FasL might play an important role in pathogenesis of viral myocarditis.

Abstract:Objective. To investigate the pathological significance of tubuloreticular structure(TRS) and cylindri- cal confronting cisternae(CCC) in patients with lupus nephritis. Methods. An electron microscopical study of 24 renal biopsy specimens from patients with lupus nephritis was carried out, with particular emphasis on two endoplasmic reticulum(ER)-related structures. Result. TRS was found in 18 cases, and CCC in 10 of them. TRS often appeared in the capillary en- dothelium, and did not correlate well with the activity index of lupus nephritis. CCC appeared frequently in monocyte/macrophage and lymphocyte, and correlated well with both the activity index and the amount of interstitial immune deposits. Conclusion.TRS and CCC derived from inward "budding" of ER membrane were suggested and the morphogenesis and morphologic variations of CCC were discussed. Both TRS and CCC are pathognomonic, though not specific changes. They may be helpful in pathologic diagnosis of lupus nephritis, when properly combined with certain clinical and pathological features.

Abstract:Sleep and waking EEG of 522 patients with epilepsy and various disease with attack nature were studied. EEG showed paroxysmal activities(PA) in 217 cases. PA appeared only during sleep in 96 cases. Posi- tive rate of EEG diagnosis increased from 23. 2 percent in waking recordings to 41. 6 percent. Fifty of 97 benign childhood epilepsy with centrotemporal spike(51. 6% ) had focal PA only during sleep. Two of 6 cases with Lennox-Gastaut syndrome showed tonic seizure and/or generalized paroxysmal fast activities, Seizure types of 15 patients were defined by interictal PA and letal EEG during sleep. There was no corre- sponding relationship between seizure time(waking or sleep) and PA sensitivity to state of vigilance.

Abstract:Objective. To investigate the peritoneum as a material using in the portal vein reconstruction. Methods. Twenty-one small healthy domestic pigs were used in the study, and divided into two groups. The observation period was twenty-four weeks, and divided into three stages. A section of portal vein about 2. 5- 3. 0 cm long was resected and replaced by autogenous peritoneum tube(APT)and autoge- nous vein respectively. The liver function tests, bloodflow dynamic examination, patency and pathological changes of the grafts were. investigaed after the replacement. Results. All animals survived the operation and the subsequent observation. It showed that aspartate transminase (AST) were mlidly elevated just after operation but recovered quickly in both groups. Portal bloodflow index(PI)and portal pressure were both a little higher than those before operations but all within normal range. Portography revealed a total patency of eight out of thirteen grafts in the experiment group, whereas a total patency of six out of eight in the control group. During the observation, portograhpy re- vealed five out of thirteen APT grafts were occluded in the experiment group, whereas two out of eight grafts were occluded in the control group. No dilatation grafts were seen. Abundant collaterals were seen around the occlusive graft. After twelve weeks of reconstruction, scanning electron microscopy revealed that APT grafts were covered with a layer of endothelial--like cells with a certain arranging direction. Conclusion. Peritoneum is a perspective self-material for portal reconstruction. Long-term patency of the APT were a little lower than that of the autogenous vein. It is worth further studying.

Abstract:Purpose. To study the interleukin 1 (IL-1)levels in aqueous humor after transscleral fixation of in- traocular lenses (IOLs) implantation in rabbits and discuss the effect of IL-1 on postoperative anterior ocular inflammation. Methods. Twenty-seven pigmented rabbits were divided into three groups: GI, transscleral fixation of posterior chamber (PC) IOLs implantation; G2, Lens of rabbits were removed without IOLs implanta- tion; G3, the control group, without surgical intervention. On the 1st, 3rd, 7th and 14th postoperative days, aqueous humor samples were obtained. Methyl thiazolyl tetrazolium (MTT) colormetry was used to detected for the presence of IL-1. The data were analyzed by using analysis of variance of SAS soft ware. Results’ It was found that IL-1 level in aqueous humor was increased after transscleral fixation of I- OLs implantation, IL-1 level reached its maximum on the 14th postoperative days in the IOL implanted group. IL-1 levels on 1st, 3rd, 7th and 14th days postoperatively was significantly higher (P<0. 05) in I- OLs implanted group than that of only extracapsular lenses extraction but no IOLs implantation group and that of the none surgical intervention group. COnclusions.IL-1 levels increased had a close relationship with a specific response to IOL implanta- tion. The increase of IL-1 may be suggested as the principal mediators of immunological and inflammatory responses, so that may play critical role in anterior ocular inflammative response after IOL implantation.

Abstract:The nitric oxide and cyclic GMP production in myocardium early after burn injury in rats were investi- gated. Nitric oxide synthase activity was measured in cytosols from the left ventricular wall of burned rats. Cytosols from the control group animals were shown to contain mainly Ca2+ dependent nitric oxide synthase (cNOS) with small amount of Ca2+ independent nitric oxide synthase (iNOS). Following burn injury, there was a marked increase in iNOS activity with a peak at sh post-burn, however, myocardial cNOS ac- tivity was found to decline obviously. Parallel to iNOS induction there was a significant increase in myocar- dial nitric oxide and cyclic GMP production. All these changes were alleviated by treatment of the rats with dexamethasone. Since increases in cyclic GMP levels in the heart were associated with reduced myocardial contractility, it is possible that enhanced production of nitric oxide by a Ca2+, independent NO synthase ac- counts, at least in part, for the depression of myocardial contractility seen in burn animals and patients.
