Abstract:On the base of the one step, operator independent method which was set up by Christophe A.E., the pancreas was infused with cold University of Wisconsin(UW) solution for the preservation, digested by the collagenase P, circuited with HBSS+5%fetal calf serum(FCS)+10mmol/L Hepes solution, and separated with the stainless steel mesh. The number of the collected islets were 400000~1800000 per pancreas, i.e. about 12150/g pancreas. After purification, the recovery was 350000~1700000 per pancreas, i.e. about 10250/g pancreas, the recovery rate was above 80%, and the purity of the final preparation was above 95%. The insulin secretion in the response to the high concentration glucose (22 mmol/L) stimulation was apparently different on the 1,3,5 day of the cultural islets, which the high level of insulin was three times the low level (5.5 mmol/L) on the 5th day, and the insulin level of the double stimulation under perfusion conditions is apparently higher than low glucose. The result demonstrated that the purified islets were functionally alive. Histological studies also show that the shape of islets are complete, and the β cell was specially stained by the dithizone (DTZ). The Trypan Blue staining had shown the living cell was above 90%. In conclusion, the new method was highly practical and yielded higher concentration of active pancreatic islets.

Abstract:The first intron of human hsp90β gene is not only essential in maintaining high constitutive expression but also critical for heat shock inducibility of the hsp90β gene. Typical HSEs in the first intron play a vital role in the heat induced expression of human hsp90β gene. Slot blot analysis shows that hsp90β gene mRNA transcripts initiated from the 3’ of the first intron dominates over that of the first exon. The intronic HSEs of the hsp90β gene show much higher binding affinity toward the recombinant heat shock factor HSF1 than that of the recombinant heat shock factor HSF2.
Abstract:The participation of immune mechanisms in the pathogenesis of dementia of Alzheimer type (AD) has been suggested. We examined T cell responses to myelin basic protein (MBP) and myelin proteolipid protein (PLP) using an enzyme linked immunospot (ELISPOT) assay by enumerating mononuclear cells (MNC) that in blood and cerebrospinal fluid (CSF) secreted the cytokine interferon γ (IFN γ) spontaneously and after short time culture of the cells in presence of MBP or PLP. These myelin components are supposed to induce autoaggressive immunity in multiple sclerosis. MBP and PLP reactive IFN γ secreting cells were detected in patients with AD and, for comparison, in patients with other non inflammatory neurological diseases(OND) and patients with tension type headache (TH). Elevated levels of MBP and PLP reactive IFN γ secreting cells were found in blood in AD patients compared to OND and TH, such cells in AD patients were further enriched in CSF. Levels of MBP reactive as well as spontaneously IFN γ secreting cells in CSF were about 180 fold and 250 fold higher than in blood of AD patients, and also higher than the corresponding data in OND(30 fold and 20 fold) and in TH (120 fold and 20 fold). It is unclear whether the autoreactive T cell responses to MBP and PLP, especially accumulated in CSF, have any importance for the pathogenesis of AD.
Abstract:Objective To detemine optimal conditions by using 5 bromodeoxyuridine (BrdU) as a marker of transplanted retinal pigment epithelial (RPE) cells in the subretinal space of albino rabbits Methods Pigmented rabbit RPE cells at second to fifth passage were fed with 20 μmol/L BrdU in Eagle’s minimal essential medium (MEM) for five days After extensive wash with phosphate buffered saline (PBS),the cells were detached by trypsin and used for transplantation onto Bruch’s membrane of albino rabbits Eyes were enucleated at various times post transplantation Acetone, 4% paraformaldehyde, periodate lysine paraformaldehyde (PLP),or half strength Karnovsky’s fixatives were individually used to fix the tissue in order to find optimal condition for detecting BrdU marker The fixation was followed by embedding in OCT compound, glycol plastic, or paraffin The transplanted area was then sectioned, pepsin digested, and used for immunohistochemical staining with monoclonal antibody against BrdU and avidin biotin alkaline phosphatase complex (ABC AP) Results Frozen sections of acetone or paraformaldehyde fixed tissue gave strong immunostaining of BrdU but the overall morphology was poor Karnovsky’s fixed tissue offered strong staining but this was buried by strong background When using PLP as a fixative, we obtained strongly positive blue staining with very low background, and also excellent morphologic preservation Conclusion In combination with immunohistochemical detection method, BrdU labeling is an excellent long term marker for RPE transplantation one year after surgery To use BrdU as a marker necessitates the use of pepsin digestion to make the BrdU antigen in the nuclei accessible to antibody But the pepsin digestion may damage other tissues and result in overall poor morphology Among the fixatives tested in this study, PLP fixed tissues offered both strong BrdU staining and good preservation of structural integrity, particularly the fine structure of photoreceptor and RPE cells
Abstract:The data from in vitro and animal experiment study has showed that costimulaory molecule B7 1 plays an important role in antitumor immunity In the present study, B7 1 expression was observed in 130 samples from a veriety of human malignancies by using immunocytochemistry, in situ hybridization and RT PCR combined with dot hybridization and B7 1 specific Mab and probe The results demonstrated B7 1 expression on tumor cells in 76 cases at both protein and mRNA level Forty two specimens were stained with B7 1 HLA ABC and HLA DR Mab and 26 showed that the three antibodies used all were positive Together with the achievement in tumor antigen study, the present findings imply that in most tumors (if not all) the tumor cells have all the requisite element to elicit anti tumor rejection response, the heterogeneous mechanism for tumor escape from immunosurvillance should be emphasized
Abstract:GBV C/HGV is a newly identified virus associated with human hepatitis In this study, the nucleotide sequences of the partial NS5 gene of GBV C/HGV derived from sera of 8 Chinese patients were determined The nucleotide homology among the 8 isolates were 92% on average On the basis of sequence analysis, two sets of oligonucleotide primers derived from highly conserved region of GBV C/HGV NS5 gene were designed to establish both sensitive and specific nested PCR for detection of GBV C/HGV RNA 253 Chinese patients were examined for the virus RNA GBV C/HGV RNA positive rates in patients infected with HBV, HCV and patients with chronic non B,non C hepatitis were 18 4%, 19 8% and 8 9% respectively This result suggested that HBV,HCV and GBV C/HGV shared the same transmission risk factors 8 patients with GBV C/HGV and HCV coinfection were retrospectively observed for the response to interferon Coinfection with GBV/HGV did not negatively influence the responsiveness of HCV, and GBV C/HGV was sensitive to interferon to a certain degree
Abstract:Objective The aim of the study was to determine whether bronchial asthma was associated with increased levels of soluble intercellular adhesion molecule 1(sICAM 1) in serum, which might be valuble data for the effective therapy of these patients Patients and methods The concentrations of sICAM 1 were determined in sera of healthy donors and asthmatic patients using a sensitive enzyme immunoassay Results The mean(±SD) levels of serum sICAM 1 of 26 asthmatic patients (205±72 0 μg/L)was significantly higher than that of the 30 healthy volunteers (154±63 9 μg/L,P<0 01) There was no much difference between the serum levels in 12 patients suffering from atopic asthma and the levels in 14 patients with nonatopic asthma The serum concentrations of sICAM 1 were higher during asthma attacks than that during remissions in the same patients (P<0 05) Conclusion These results suggest that sICAM 1 may play a certain role in the pathophysiology of bronchial asthma, and might be signals for successful treatment
Abstract:A total of 19 cases were performed the resection of the body and tail of the pancreas with preservation of spleen in our department since it was first successfully carried out in the late 1980′s The indications of operation were pancreatic cystadenoma(n=8),pseudocyst(n=2),insulioma(n=3),pancreatic injury(n=6) All but one went well intraoperatively and postoperatively with the normal appearance and function of the spleen The main observation items of postoperation were leukocyte phagocytosis test, splenic CT scan, ultrasonography and Technetium 99m spleen scan The exceptional one showed transient insufficiency of splenic blood supply after the operation Considering the regional conditions in the operations, the procedure could be classified into the resection of the body and tail of pancreas with and without conservation of the splenic artery and vein
Abstract:Identification of Cryptococcus neoformans(C neoformans) has been considered depends on the phenotype and biochemical characteristics, especially on its urease positive characteristics During the last ten years, there are 4 strains of urease negative C neoformans strains have been discovered, of which 2 were isolated from AIDS patients, 1 from pigeon dropping and another 1 from preserved cultures of 286 strains of C neoformans So far, from medlines, there was none of urease negative strain isolated clinically from non AIDS patient was report Recently, we isolated a strain of urease negative strain of C neoformans from neither AIDS nor immunocompromised patients for the first time
Abstract:In order to investigate the differences of cellular electrophysiologic changes between laser photocoagulation(LP) and radiofrequency ablation(RFA), transmembrane potentials were recorded at 0 5,1,3,5,8 mm from lesions before and after LP and RFA had been performed respectively on 15 endocardial tissues removed from 15 adult rabbits in each group Resting potential(RP), action potential amplitude(APA), maximum rate of upstroke velocity (dV/dT), and action potential duration at 50 percent of repolarization(APD50) decreased at 0 5, 1,3 mm from lesions in RF, so did all of them at 0 5,1 mm in LP, but all transmembrane potential variables were almost normal at 3 mm in LP Furthermore, almost all the transmembrane potential variables at 0 5 1,3 mm in RFA went down much lower than those in LP(P<0 01) There was no difference in all transmembrane potential variables at 5, 8 mm between LP and RFA APD 90 had no difference at varing distance in both the groups Electrophysiologic abnormalities of border zone of injured but nonnecrotic myocardium around the lesions of laser photocoagulation are much less than those of radiofrequency ablation in strength and much smaller in scope, which suggest that unfavorable effects of laser photocoagulation on hemodynamics and arrhythmogenicity would be less than those of radiofrequency ablation
Abstract:The CT including HRCT appearances of six patients with histopathologically confirmed pulmonary alveolar proteinosis(PAP) were evaluated and compared with those of chest radiographs In all patients the CT manifestations were quite similar: bilateral and diffuse airspace consolidation was usually patchy or confluent with sharply defined margins, intermingled with normal lung tissue The configuration of lung lesions was “geographical” in outline with angulate, strait and curved margins There were white branching linear opacities within the ground glass background Although various pulmonary diseases may mimic PAP in some way, a full awareness of the characteristic CT appearances of PAP is helpful in achieving a correct diagnosis CT may provide more accurate evidence than chest radiograph for the evaluation of the extent and delineation of PAP
Abstract:Method Fifteen hundred and fifty two patients with heart disease were examined with monoplane, biplane and omniplane TEE probe(including male 727, female 825;9~76 years old) During the same period, 113 307 precordial echocardiographic examinations were performed in our laboratory The ratio between TEE and transthoracic Echo examines was 1:73 Result All different kinds of complication were occurred during TEE examination, including vomiting, minor mucus bleeding, laryngospasm, mandibular dislocation, angina pectoris, arrhythmia, even ventricular fibrillation and death The article suggested that there were five steps must be improved during TEE examination including instrument and patients preparation, local anesthesia, manipulation technique and TEE probes pattern Conclusion Although TEE is a semiinvasive technique and can cause some kinds of complication, it is a safe technique if the five steps are improved
Abstract:Objective. To investigate the role of amputation in management of severely injured extremities and factors that may influence decision making. Methods. Thirty six patients with amputations following severe injuries of the lower extremities were retrospectively reviewed. Results. There was one death from multiple organ failure. Among the remaining 35 cases, primary and delayed wound healing was achieved in 22 and 13 patients, respectively. Conclusion. Amputation is an important means for management of severely injured lower extremities. When salvage is unlikely to lead to the functional reconstruction of the limb, amputation should be indicated.
Abstract:The polymorphism of apoE, an important determinant in the lipid metabolism, may be related to the atherosclerotic events. In this study, the apoE genotypes, by means of PCR AFLP assay in 42 patients with cortical cerebral infarction and 85 healthy controls were detected. The results showed that the apoE ε 3/3 genotype was more prevalent in the controls (55.29%), and ε 3/4 genotype was more common in the patients (57.14%). The frequency of ε 4 allele was more predominate in the patients than that in the controls. The ε 3/4 genotype or ε 4 allele was associated with 2.44 or 2.31 fold increase in the risk of cortical cerebral infarction. Therefore, it was suggested that apoE ε 4 allele is a genetic factor of susceptibility to cerebral infarction, which supported the hypothesis of the association between apoE polymorphism and atherosclerotic cardio cerebrovascular disorders.
